Johns Hopkins researchers link reversible 'epigenetic' marks to behavior patterns

Johns Hopkins scientists report what is believed to be the first evidence that complex, reversible behavioral patterns in bees and presumably other animals are linked to reversible chemical tags on genes.

The scientists say what is most significant about the new study, described online September 16 in Nature Neuroscience, is that for the first time DNA methylation "tagging" has been linked to something at the behavioral level of a whole organism. On top of that, they say, the behavior in question, and its corresponding molecular changes, are reversible, which has important implications for human health.

According to Andy Feinberg, M.D., M.P.H., Gilman scholar, professor of molecular medicine and director of the Center for Epigenetics at Hopkins' Institute for Basic Biomedical Sciences, the addition of DNA methylation to genes has long been shown to play an important role in regulating gene activity in changing biological systems, like fate determination in stem cells or the creation of cancer cells. Curious about how epigenetics might contribute to behavior, he and his team studied a tried-and-true model of animal behavior: bees.

Working with bee expert Gro Amdam, Ph.D., associate professor of life sciences at Arizona State University and the Norwegian University of Life Sciences, Feinberg's epigenetics team found significant differences in DNA methylation patterns in bees that have identical genetic sequences but vastly different behavioral patterns.

Employing a method that allows the researchers to analyze the whole genome at once, dubbed CHARM (comprehensive high-throughput arrays for relative methylation), the team analyzed the location of DNA methylations in the brains of worker bees of two different "professions." All worker bees are female and, within a given hive, are all genetically identical sisters. However, they don't all do the same thing; some nurse and some forage.

Nurses are generally younger and remain in the hive to take care of the queen and her larvae. When nurses mature, they become foragers that leave the hive to gather pollen and other supplies for the hive. "Genes themselves weren't going to tell us what is responsible for the two types of behavior," Feinberg says. "But epigenetics and how it controls genes could."

Feinberg and Amdam started their experiment with new hives populated by bees of the same age. That removed the possibility that any differences they might find could be attributed to differences of age. "When young, age-matched bees enter a new hive, they divvy up their tasks so that the right proportion becomes nurses and foragers," explains Amdam. It is these two populations that were tested after painstakingly characterizing and marking each bee with its "professional," or behavioral, category.

Analyzing the patterns of DNA methylation in the brains of 21 nurses and 21 foragers, the team found 155 regions of DNA that had different tag patterns in the two types of bees. The genes associated with the methylation differences were mostly regulatory genes known to affect the status of other genes. "Gene sequences without these tags are like roads without stop lights gridlock," says Feinberg.

Once they knew differences existed, they could take the next step to determine if they were permanent. "When there are too few nurses, the foragers can step in and take their places, reverting to their former practices," says Amdam. The researchers used this strategy to see whether foraging bees would maintain their foraging genetic tags when forced to start acting like nurses again. So they removed all of the nurses from their hives and waited several weeks for the hive to restore balance.

That done, the team again looked for differences in DNA methylation patterns, this time between foragers that remained foragers and those that became nurses. One hundred and seven DNA regions showed different tags between the foragers and the reverted nurses, suggesting that the epigenetic marks were not permanent but reversible and connected to the bees' behavior and the facts of life in the hive.

Dramatically, Feinberg noted, more than half of those regions had already been identified among the 155 regions that change when nurses mature into foragers. These 57 regions are likely at the heart of the different behaviors exhibited by nurses and foragers, says Amdam. "It's like one of those pictures that portray two different images depending on your angle of view," she says. "The bee genome contains images of both nurses and foragers. The tags on the DNA give the brain its coordinates so that it knows what kind of behavior to project."

The researchers say they hope their results may begin to shed light on complex behavioral issues in humans, such as learning, memory, stress response and mood disorders, which all involve interactions between genetic and epigenetic components similar to those in the study. A person's underlying genetic sequence is acted upon by epigenetic tags, which may be affected by external cues to change in ways that create stable but reversible behavioral patterns.

Female astronaut takes command of space station

NASA astronaut Sunita Williams, who holds the record for the longest spaceflight by a woman, took charge of the International Space Station Saturday, becoming only the second female commander in the orbiting lab's 14-year history.

Williams took charge of the space station from Russian cosmonaut Gennady Padalka, who is returning to Earth on Sunday after months commanding the outpost's six-person Expedition 32 crew. Williams launched to the station in July and will command its Expedition 33 crew before returning to Earth in November.

"I would like to thank our [Expedition] 32 crewmates here who have taught us how to live and work in space, and of course to have a lot of fun up in space," Williams told Padalka during a change of command ceremony. She will officially take charge of the station on Sunday, after Padalka and two crewmates board their Soyuz spacecraft for the trip home.

Padalka, NASA astronaut Joe Acaba and cosmonaut Sergei Revin are scheduled to undock from the space station Sunday at 7:09 p.m. EDT (2309 GMT) and land in the Central Asian steppes of Kazakhstan at 10:53 p.m. EDT (0253 on Sept. 17). The trio is wrapping up a five-month mission to the space station and Padalka thanked his crewmates and flight controllers on the ground for their help during the flight.

Sunita Williams arrived at the space station on July 17 on a Soyuz spacecraft with two crewmates: Japanese astronaut Akihiko Hoshide and Russian cosmonaut Yuri Malenchenko. They will be joined by three new crewmembers in October.

Williams, 46, is a captain in the U.S. Navy and flying on her second long-duration space mission. She first launched into space in 2007 and spent 195 consecutive days in space, setting a record for the longest single spaceflight by a female astronaut. On Sept. 19, she'll celebrate her birthday in space.

In a NASA interview before launch, Williams said a friend asked her if she was nervous about commanding the space station. She said no, adding that the more than two years of training alongside her Expedition 32 and 33 crewmates, as well as the Mission Control team, prepared all the space station crewmembers for life in space.

"When you get up on the space station, you know what to do, so I?m not nervous about it all," Williams said. "I?m psyched."

The first female commander of the International Space Station was NASA astronaut Peggy Whitson, who commanded the orbiting lab's Expedition 16 mission between fall 2007 and spring 2008. Whitson later served as NASA's chief astronaut and recently stepped down from the post to rejoin the Astronaut Corps' spaceflying ranks. Two other female astronauts, Eileen Collins and Pam Melroy, commanded NASA space shuttles.

Mission Control sent Williams a few words to mark her new role as space station skipper.

"Congratulations on the promotion to your new position," a flight control told Williams after today's ceremony. "We know you'll run a fine ship up there."

Chinese protesters clash with police outside Japanese embassy ( video)

People are in the streets over a chain of islands disputed by China and Japan; the issue bubbled to the surface last week when the Japanese government purchased the islands from their private owner.

Protests against Japan over its control of disputed islands spread across more than two dozen cities in?China?and turned violent at times Saturday, with protesters burning Japanese flags and clashing with Chinese paramilitary police at the Japanese Embassy before order was restored.

Thousands of protesters gathered in front of the embassy in Beijing. Hundreds tried to storm a metal barricade backed by riot police armed with shields, helmets and batons. Many threw rocks, bottles, eggs and traffic cones at the embassy.

The embassy said protesters around the country set fire to Japanese factories, sabotaged assembly lines, looted department stores and illegally entered Japanese businesses.

"We express regret over what has happened today and ask the Chinese government to ensure the safety of Japanese citizens and businesses in?China," it said in a statement.

Anti-Japanese sentiment, never far from the surface in?China, has been building for weeks, touched off by moves by Tokyo and fanned by a feverish campaign in Chinese state media. Passions grew more heated this past week after the Japanese government purchased the contested East?China?Sea islands from their private Japanese owners.

Japan's Kyodo News agency said more than 60,000 people protested in at least 28 Chinese cities, making the anti-Japanese demonstrations the largest since the two countries normalized diplomatic relations in 1972. The protests were expected to continue Sunday.

Although Japan has controlled the uninhabited islands ? called Diaoyu in Chinese and Senkaku in Japanese ? for decades,?China?saw the purchase as an affront to its claim and as further proof of Tokyo's refusal to negotiate over them.

Beijing made angry protests and tried to bolster its claim by briefly sending marine surveillance ships into what Japan says are its territorial waters around the islands and by ratcheting up state media coverage. Some news programs featured bellicose commentary.

In Japan, candidates vying to lead the top opposition party called for a tough stand against Beijing in the dispute.

Shigeru Ishiba, a former defense minister seen as a leading contender to head the Liberal Democratic Party, said in an election debate that Japan should send a strong message to?China?that it will not back down.

"This is something that Japan should do as a nation," he said.

Smaller demonstrations had been staged in?China?throughout the week. But they boiled over Saturday, especially in Beijing. Outside the Japanese Embassy, the protesters ? most of whom appeared to be students ? waved Chinese flags and shouted slogans demanding that Japan relinquish the islands. Some hurled rocks, bottles and traffic cones at the embassy. As the crowd grew, police closed off a main thoroughfare to traffic. City buses skipped the stop near the embassy.

Zhang Zhong, a 32-year-old computer worker, said Chinese should stand up against Japan, remembering its brutal occupation of much of?China?before and during World War II.

"We cannot lose the Diaoyu Islands," he said. "We cannot forget our national shame."

A protest in Shanghai was orderly. About 200 police officers cordoned off the street leading to the Japanese Consulate, allowing protesters in groups of 100 to approach the building. Demonstrators had to register first with police.

But in Changsha, protesters ransacked the Japanese department store Heiwado. They also smashed a police car made by Mitsubishi and overturned another Japanese car, according to online reports. Provincial police asked motorists driving Japanese-brand cars to avoid major thoroughfares and refrain from parking on the street.

Kyodo said protesters ransacked at least 10 Japanese restaurants in Suzhou and damaged a Jusco supermarket run by Japan's Aeon group in Qingdao.

Li Yiqiang, a Chinese activist for the islands, said he opposes violence but that heated behavior is unavoidable when strong feelings boil over.

"When the national emotions erupt, it is understandable that some people would overreact," Li said. "How can you control spontaneous acts?"

The demonstrations came before an anniversary Tuesday of the 1931 Mukden Incident which often triggers anti-Japanese sentiment. The incident was used by Japan as a pretext to invade northern?China, and activists have called for more demonstrations on that day.

The Japanese government had hoped its purchase of the disputed islands would calm rather than inflame the situation. The nationalistic governor of metropolitan Tokyo, Shintaro Ishihara, had proposed buying the islands in April and planned to develop them ? something that Beijing would have seen as a powerful attempt to solidify Japan's claim. By purchasing them itself, the central government promised to keep them undeveloped.

Sorry, Big Tobacco, Norway Doesn't Like You

This hasn?t been the best year for tobacco companies. The Centers for Disease Control and Prevention announced more graphic anti-smoking ads, Australia kicked tobacco company logos off cigarette packs, and now a Norway court has upheld a ban on tobacco store displays.

The Norway ruling is the latest blow to Big Tobacco, which is working hard to maintain its presence around the world as anti-smoking campaigns and legislation try to thwart its efforts.

Norwegians have banned cigarette and alcohol advertising since 1975, the Associated Press reported, and in 2010 the government said no to tobacco store displays.?Philip Morris challenged that law with a suit, and with this loss the company now may appeal the decision.

Reuters reported that the country might follow Australia?s example and not allow tobacco companies to display their logos on cigarette packages.

"I know that this ruling will be read carefully in other European countries," Knut-Inge Klepp, director at the Norwegian Health Directorate, was quoted as saying. "Currently we are waiting for a new strategy from the government on tobacco legislation. We are in dialog with other European countries on the issue.?

Australia?s mandate to replace cigarette package logos with graphic photos of rotted teeth and blinded eyeballs is considered the strictest tobacco labeling law in the world. That, too, was challenged by tobacco companies, which lost the fight.

But elsewhere in the world cigarette companies have a stronger foothold. A recent study in the Lancet on smoking rates around the world found that about 41 percent of men who live in developing countries are smokers. Advertising campaigns in some of these countries tout smoking as glamorous and good for you.

Karl Erik Lund, a research director at the Norwegian Institute for Alcohol and Drug Research told AP, ?This verdict sends a signal that it's possible to win over the mighty tobacco industry.?

A couple of weeks ago, I had the pleasure of interviewing cookbook author Norene Gilletz. Norene lives in Toronto, Canada. She?s the author of several well known cookbooks, including The New Food Processor Bible, Norene?s Healthy Kitchen, Healthy Helpings, and several other dietary cookbooks. Norene is well known for her healthy Jewish and kosher cooking; her site Gourmania.com is a popular resource for recipes, cooking advice, and more. She also has a fun cooking group online called Norene?s Kitchen, which is where I first ?met? her online. I had a great time speaking with her on the phone and getting to know her better. My mother was born in Russia. My father?s family came from Poland.

How long ago?

My mother came when she was 6, so in about 1921. My father was born in Winnipeg. My mother passed. She?d be 98 today, so almost 100 years. She never needed a recipe. She could cook anything.

When you were growing up and celebrating Rosh Hashanah with your family, what were your favorite dishes that your mom made?

My mother always made potato knishes and carrots and honey cake. Some sort of chicken. The joke was, well my mother didn?t really go to shul, she sent us with my father. So my mother would say, ?your bubbe is going to ask you what I made, so tell her that I made roast chicken.? And I?d say, ?but Mama, you didn?t, how can I tell a lie?? And she?d say that my grandmother would expect to hear that she made roast chicken, so tell her that. She?d always make a carrot dish that I have the recipe for somewhere. She made honeycake with buckwheat honey, because it had a different flavor. And she always made her honeycake with tea, not coffee. I remember the buckwheat honey was very thick. And I don?t really have her recipe, I just take my recipe and modify it. I remember in my mind how my mother made it.

So would you say your mom was a major influence on your culinary career.

Absolutely. Without a doubt.

Norene, you?re the master of the food processor. Your book,?The Food Processor Bible, was recently released in a 30th?anniversary edition. How did you get into the whole food processor thing?

Years and years ago, I was part of a committee that did a fundraising cookbook called?Second Helpings Please. We were all in our 20s, and we were really trying to capture our mother?s and our grandmother?s recipes to put them into print. We thought it would take us about 3-4 months to collect everything, and it took about 3.5 years. So we worked hard to promote the book, and as a result people would call me asking how do you make this, how do you do that? so I was cooking and baking a lot. Then I went to a trade show and saw the food processor in action. I had started writing my own cookbook, everyone was saying ?you should do something yourself,? and I felt I could. I had lots of energy and nobody could keep me down. So by then I had done about half a book. You know, all of my favorite kinds of recipes? and I met my first food processor. After about a month of using the machine, I thought, you know what? I?ve got this thing mastered, and I can do things quickly and efficiently, and it just saved time. My mother was always looking for shortcuts. She had a little hand grater, she had a hand-turning-thing where you would change the disc, almost like a meat grinder. I forget what they called it. But suddenly here was this food processor, and the results were fabulous. And the machines were so simple to use. So I took all of my recipes and converted them for the food processor.

I?m curious about what drew you to a career in cooking.?

When I was working on the fundraising book, I leaned that I could explain recipes very clearly, so that people could reproduce them. I had taught swimming when I was in my teens, so I was very good at presenting visual images. In my own learning pattern, if I would have a visual image, then it was very easy for me. Everything I write has a visual image, because otherwise I don?t understand the instructions myself. Because of that, my instructions are very clear. The deficit made me a much better writer. And I love food. I was very fascinated by how things were made. Everything was new. I had an instinct for a good recipe; I just knew it was good. I knew how to capture it and write it down. There were no computers then. When we did Second Helpings, we typed the recipes on 4 x 6 cards. And then we made a binder, and I kept moving things around. You know the little photo holders? Every time we got a new recipe, I kept having to move everything all the way down to add to a section. I love to teach and share. I was always bringing things to my kids? school, and I was very involved the community. I cooked all the time and I was brought up that you prepared it at home. My mother would come to visit or I would go to Winnipeg and watch how she made things.

When did you decide that food would be your life?s work?

I joined the IACP. There were no cooking classes available in Montreal. I went to an IACP conference, and I met people who understood me. My friends thought I was crazy because all I was interested in talking about was how you make this thing or that. And when I went to IACP, everybody understood what I was about. I had people I could talk to. I wasn?t interested in playing games or tennis. That just didn?t ring my chimes at all. I love the art of creating. You know, I watched Julia Child, learned how to make puff pastry. I used to go in the kitchen after my kids were asleep and cut onions different ways, and I would make onion blooms with different garnishes. I?m left handed, so I had challenges in learning how to do things that were pretty or fancy. I?m not a fancy cook, but I write very clearly. If I give you a recipe, it works. My conscience knows it?s right. When I try other people?s recipes, I know the error. I just know. I create a visual image when I write, when I edit my own work or anybody else?s work, and I can find an error in a book like you wouldn?t imagine.

You obviously love to teach and share your cooking knowledge. I?ve noticed that some people are protective of their recipes. Can you relate to that at all? Do you have a few recipes that you will never share because they?re so special, or are you an open book?

I always share, and I?ll tell you why I share. A woman told me a story about a dozen years ago about her father?s bakery in Winnipeg, which is where I grew up. And I remember going to that bakery as a little girl and I remember the smell of everything, you know that wonderful yeasty smell? I?d go in with my mother and it was such a good sensation to be able to smell everything? the warm baked goods on the walk home. So, she told me her father owned the bakery, and his head baker had a bread recipe that he would never share? and then he died. What?s the point of knowing something? I know some people have these secret ingredients that they would never share. Like my aunt made these amazing cinnamon buns, and she used real butter, and her technique with the yeast was so incredible. She could pull dough so thin you could really read the newspaper through it. And I would tell my aunt her cinnamon buns were so good, and my mother would ask what was wrong with her cinnamon buns. Like it was a competition. So people cooked their trademark dish. And there was an issue with people taking their recipe and turning it into a commercial product. It?s an issue of intellectual property. You know, it doesn?t matter what the recipe is., if someone gives you a recipe there?s always something missing or changed. I don?t do that, though. I like sharing.

I always think of recipes as being short stories. And people often do share. They?re looking for a taste memory, because a memory of food will literally transport you back to that table as a child or whenever. There?s a group called Montreal Memories and they go into these raptures about food they remember from their childhood. I remember cherries from when I was 3 or 4 years old with my zayde (grandfather). I don?t remember much else, but that he smelled like tobacco and the cherries. The cherries bring back a strong memory. Me sitting on the grass and wiggling my toes. It?s an old one.

If you could pick one food that?s your favorite in the world, what would it be?

Kasha. I love the fried onions. I usually cook one box of kasha with one egg. I love the aroma. I?ll double the amount of liquid. And lots and lots of fried onions that are nice and crispy golden brown. I cook them in olive oil, and sometimes I put mushrooms in. I?ll make a meal from it. Kasha and bowties, or any type of pasta that I might have. My bubbe used to make handmade dough. She would make a pasta dough, roll it out and cut it into squares and boil it very quickly. And I like matzo balls. I like all of my old traditional stuff. My tastes have changed. I can?t eat a lot of the heavy stuff anymore. I?ll have a steak once in a while. I like a good salami sandwich, but not very often. Growing up we always had a dried salami hanging in the closet next to the broom. Now I can?t eat it the same way because of all of the salt. It?s a treat. I eat a lot of vegetarian. I love a dairy kugel or a vegetable kugel. I like potato latkes. And I love to share food with friends.

After interviewing Norene, I decided to make a recipe from one of her most popular cookbooks, The Food Processor Bible. While the recipe isn?t specifically a Rosh Hashanah dish, it incorporates sweet, natural flavors like carrot, sweet potato, and honey. It?s very low fat and healthy (like many of Norene?s recipes), with a lovely garden-fresh flavor from the addition of fresh dill and basil.?Norene says:

?This easy, nutritious soup is packed with beta-carotene. You can use a combination of carrots and squash for a tasty variation.?

This soup would make a nutritious vegan addition to the Rosh Hashanah menu. It?s also a great option for Sukkot because of the beautiful autumn color and the inclusion of seasonal root vegetables. Enjoy!

Kosher Key: Pareve

Ingredients

  • 2 cloves garlic
  • 1 slice fresh ginger, peeled (about 1 tbsp)
  • 1 large onion, cut in chunks
  • 1-2 stalks celery, cut in chunks
  • 1 tbsp olive oil
  • 1 1/2 lbs carrots, cut in chunks (4 cups)
  • 2 medium sweet potatoes
  • 1 medium potato
  • 8 cups water or vegetable broth
  • 2 tsp salt (or to taste)
  • Freshly ground black pepper to taste
  • 2 tbsp fresh basil
  • 2 tbsp fresh dill
  • 1-2 tbsp fresh lemon juice (or to taste)
  • 1-2 tbsp honey (or to taste - omit to make soup vegan)
  • Sprigs of fresh dill for garnish (optional)

You will also need

  • Food processor, large soup pot, immersion blender (optional- recommended)

Prep Time: 20 Minutes

Cook Time: 35 Minutes

Total Time: 55 Minutes

  • Place the steel blade on your food processor. Drop garlic and ginger through feed tube while machine is running.
  • Process until minced. (Use mini-bowl and mini-blade if available) Set aside.
  • Process onion and celery with quick on-off pulses, until coarsely chopped.
  • Heat oil in large soup pot. Saute onion and celery for 6 or 7 minutes, until tender, stirring occasionally. Add a little water if the vegetables begin to stick.
  • Process carrots with quick on/off pulses, until coarsely chopped.
  • Add to soup pot along with reserved garlic and ginger; cook 2 minutes longer.
  • Coarsely chop potato and sweet potatoes. Add to soup pot along with water or broth, salt and pepper.
  • Bring to a boil, reduce heat and simmer, partially covered, for about 30 minutes, until vegetables are tender.
  • Process basil and dill until minced, about 10 seconds. Reserve.
  • When the vegetables are nice and tender, puree the soup. Because of the quantity, it is easiest to use an immersion blender and puree the soup directly in the pot.
  • Alternatively, to puree soup in processor, place a strainer over a large bowl or saucepan. Strain soup, reserve cooking liquid. Puree solids on the steel blade attachment until smooth, working in batches if necessary. Stir pureed vegetables into the reserved cooking liquid.
  • Add minced basil, dill, lemon juice and honey to the soup (I used the immersion blender to blend them into the puree). Season with salt and pepper to taste. Add additional water if the soup is too thick.
  • Serve hot. Garnish with a sprig of fresh dill, if desired. Keeps 3 days in the refrigerator. Reheats and/or freezes well.

All of the grid patterns are knotted in the same way- hold the pattern horizontally for letters that run across the bracelet or hold the pattern vertically for keychain letters... either way you just follow the grid and count the boxes. It's both identical you just need to COUNT! ____________________________________________ When making any Alpha bracelet, letters or pictures, you are working like a typewriter. You weave a foreground string back and forth and add knots from the background in order to achieve a pattern. When you're bringing the background strings to the foreground (adding the lettering) make sure to keep the original foreground string fairly loose as you knot with the background strings so you don't flip your knots. Keeping the foreground (green) string too taught will make the (pink) background strings pull away from the previous row. ______________________________________________________ All alpha patterns are based off of and created with grids. Work one horizontal row at a time going forward for the first row, backward for the second, forward for the third and so on. Just go back and forth and count/mimic the boxes in order of appearance on your grid. ? In a forward knot row you need to knot the background strings BACKWARD onto the foreground string. ? In a backward knot row you need to knot the background strings FORWARD onto the foreground strings. ?
